図書館 - is an entire building that is a library,
図書室 - is a room within a building that is a library
The first is the library building, the second is a library room

If you look at the last kanji of each of them you will notice one means room and the other means building. The first two Kanji when put together mean books. So basically you have a book building(library) and then a book room(library room).
Library (room) is 98% of the time in a school.
