Yes, there should be a の between 山 and 手.
ya it is a bummer that they don't include the の for the train line name.- Well, the good part is that only the correct answer has the 線 kanji in it so if you know to search for that fist you won't make a mistake again.

eh, you get used to it. One off reads for Jukugo are all over Japan. Always trainlines and locations. Its just part of the Japanese language and culture. The way one is read in Tokyo will be completely different than its read in Osaka or Nagoya. This and Kanji that are no longer used come up all the time, just focus on remembering the Jukugo, especially if you will be needing that trainline (How i recognize the 山手線 lol)
